Romney: American kids get 'third-world education'
WASHINGTON (AP) ? Charging that American students are getting a "third-world education" under President Barack Obama, Republican presidential challenger Mitt Romney on Wednesday proposed a voucher-style system that could significantly alter the public school system and revive the debate over school choice.
Romney, who has been reluctant to stray far from the economic issues at the core of the presidential campaign, outlined the proposal during a speech Wednesday at the U.S. Chamber of Commerce.
"Millions of kids are getting a third-world education. And America's minority children suffer the most," Romney declared. "This is the civil-rights issue of our era. And it's the great challenge of our time."
He continued: "President Obama has made his choice, and I have made mine. As president, I will be a champion of real education reform in America."
Romney said he would let low-income and disabled students use federal money to attend public schools, public charter schools and, in some cases, private schools. Federal funds could also be used for tutoring or digital courses.
The plan is line with GOP reforms aimed at giving students more educational choices. But it's unclear how schools in areas that depend on the federal funding would fare.
The proposal was not expected to include any new federal money for education.
Romney so far has offered few details for his plans on several key policy areas, including foreign policy, health care and education. He attacked Obama's education policy and said it's heavily influenced by campaign contributions from teachers' unions.
"The teachers' unions are the clearest example of a group that has lost its way," Romney said. "The teachers unions don't fight for our children."
The message is consistent for Romney, who regularly heaps criticism on the Democratic president's policies but until now has only offered a vague road map for what he would do as president.
White House spokesman Jay Carney said Wednesday that Romney's shift to education was welcome after a campaign season in which he said the GOP rarely mentioned the issue.
"Education never came up in the Republican primary in any of the debates, or if it did, it came up almost never," Carney said.
Carney said Obama's education initiatives have received broad bipartisan support and that the president "looks forward to defending that record."
Taking a sharper tact, Obama's campaign said the budget Romney signed into law as Massachusetts governor cost 14,500 teachers, librarians and school police officers their jobs.
"Not exactly the record of a job creator," campaign spokesman Ben LaBolt said.
"Mitt Romney gave a vague, detail-free speech this morning on education and confirmed, as if there were any doubt, just how far back he would take us," LaBolt told reporters on a conference call arranged to respond to Romney's speech.
Romney's shift carries some risk. His regular criticism of labor unions, in particular, threatens to alienate voters in Rust Belt states like Michigan, Ohio and Pennsylvania, where a close election may be decided.
Before the speech, Romney announced Tuesday a team of education policy advisers that includes former Education Secretary Rod Paige and other officials from President George W. Bush's administration. Paige is among several prominent opponents of teachers' unions on the panel. As education secretary in 2004, he labeled the National Education Association a "terrorist organization."
Romney's positions on education have evolved over time. He once supported abolishing the Education Department but reversed that position as a presidential candidate in 2007. At the time, he said he came to see the value of the federal government in "holding down the interests of the teachers' unions" and putting kids and parents first.
Romney also changed his position on the Bush-era education overhaul known as "No Child Left Behind." He said he supported the law as a candidate in 2007, but he has since generally come out against the policy many conservatives see as an expansion of the federal government.
Romney continues to support the federal accountability standards in the law, however. He also has said the student testing, charter-school incentives and teacher evaluation standards in Obama's "Race to the Top" competition "make sense," although the federal government should have less control over education. The campaign in recent days has emphasized his support for charter schools while governor of Massachusetts, a theme Romney weaved into Wednesday's address and met with opposition from Obama's team.
"We know from experience that private school vouchers have failed to raise achievement and they drain resources from public schools that serve the vast majority of children," Obama policy director James Kvaal said on the conference call.
The speech represents Romney's first public event in four days. Working to close Obama's cash advantage, he's coming off a three-day fundraising swing in the New York area that his chief finance aide said had netted $15 million. A single finance event in Manhattan on Tuesday evening generated $5 million.
Still, the campaign is eager to drive a positive message for voters now tuning in to the contest.
The education speech follows a relatively quiet phase for Romney, who has been focused on fundraising but usually delivers one major address a week. Most of his recent speeches, however, have been about the economic themes that so far have defined his campaign.

Prostate Cancer Screening Dies a Quiet Death
?It?s official,? says ConsumerReports.org, ?The PSA blood test to screen for prostate cancer has been downgraded to the lowest possible rating of ?D? from the U.S. Preventive Services Task Force,?an influential independent advisory board that basically decides what types of preventive care are worth the bother.? The task force is advising physicians to discourage patients from taking the test.?
For many people that?s probably a relief. For me, the ?D? stands for ?dismayed.? Yes, I know that most prostate cancers grow so slowly that affected men will most likely die of something else first. And Consumer Reports points to results from the clinical trials indicating, ?between zero and one person will avoid death from prostate cancer in a decade for every 1,000 people that get the test.?
Yet I also vividly recall the day when cancer came calling. I was caught off guard since I was only 26 years old and had always been blessed with good health. But after experiencing recurring bladder infections that were only temporarily cured with antibiotics, a doctor recommended I have a cystoscopy.
Even a description of this mercifully quick procedure is not for the faint of heart: Essentially, a scope was inserted into my urethra and slowly advanced into the bladder. By the way, I was completely awake and not under any form of anesthesia when this occurred.
During that first exam, the urologist discovered that I had a very small malignant bladder neoplasm. It was removed using tiny surgical instruments that can be inserted through the cystoscope. I was lucky that day and my luck has held out. I?ve continued to have an annual cystoscopy for many years with no signs of recurrence.
Given the close relationship between the bladder and the prostate, part of my exam always included a PSA blood test. It was my extra bit of reassurance that everything really was OK?I even kept my only little log of the results (.35 in November 2011).
However, now it?s official. My PSA security blanket has been taken away from me. I?m sure I?ll adjust to this new reality. But like a lot of other things we don?t like to give up, it?s going to take some getting used to.
How do you feel about the Preventive Services Task Force rating?

Distress of child war and sex abuse victims halved by new trauma intervention

Queen's University Belfast
A new psychological intervention has been shown to more than halve the trauma experienced by child victims of war, rape and sexual abuse.
Researchers at Queen's University Belfast pioneered the intervention in conjunction with the international NGO, World Vision as part of a wider programme to treat psychological distress in child victims of war and sexual violence in the Democratic Republic of Congo (DRC).
Eastern Congo has the world's highest rate of sexual violence. Known as 'the rape capital of the world', it is estimated that girls and women in the eastern DRC are 134 times more likely to be raped than their counterparts in the West.
After only 15 sessions of the new group-based Trauma-Focused Cognitive Behaviour Therapy (TF-CBT), Queen's researchers found reductions of:
- 72 per cent in trauma symptoms in female victims of rape and sexual abuse
- 81 per cent in feelings of depression and anxiety
- 72 per cent decrease in conduct disorder
- 64 per cent decrease in anti-social behaviour
In war-affected countries, such as the DRC, victims of rape and sexual violence often do not receive any psychological or even medical help.
In the Queen's study, the children received sessions of trauma psycho-education, relaxation techniques, mental imagery techniques, and tips on how to identify and change particular inaccurate or unhelpful cognitions. The girls also drew pictures of their most traumatic events and were encouraged to talk about these events in individual sessions with Queen's psychologists and a team of Congolese counsellors who were trained by the University's lead researchers, Paul O'Callaghan and John McMullen.
Speaking about the implications of the results for treating child victims of war and sexual abuse worldwide, Paul O'Callaghan, from Queen's School of Psychology said: "In war-affected areas, victims will often not seek, or receive help of any kind. This is because wives fear that their husbands will reject them, and girls fear that they will be unable to marry if it is known that they have been violated. Stigma is also compounded by the fact that many of these rapes are carried out by undisciplined militias operating in remote and inaccessible parts of the country and the nearest hospital or clinic is many miles away.
"It is not surprising that studies show sexual abuse to have a profoundly detrimental effect on the mental health of girls in war-affected countries, but what is surprising was just how successful the intervention was in reducing psychological distress.
This is the first time that such a therapy has been used in this way. Although Trauma-Focused Cognitive Behaviour Therapy has the greatest evidence for its effectiveness in treating child victims of sexual violence in the West, to date no study has ever adapted it for use in developing countries affected by the twin evils of war and sexual violence.
"The dramatic reduction in trauma, depression and anxiety, conduct problems and anti-social behaviour shows that this kind of therapy is very effective in treating war-affected children who have been exposed to rape and sexual violence. In addition to the statistical results of the therapy many of the girls attested to how the intervention helped reduced their terrible nightmares, disturbing flashbacks and suicidal thinking. For me, that was the most rewarding part of our work in the DRC."
The study, which took place over five weeks in 2011, also treated the psychological distress of 50 war affected boys between the ages of 12 and 17. It was shown to dramatically reduce levels of trauma, depression and anxiety, conduct disorder and anti-social behaviour in male child soldiers and street children.

2. There are about 300,000 child soldiers in the world today and nearly, one in every three is a girl. Girl soldiers are at greater risk of psychological damage due to the high rates of sexual violence in war. A recent study in the DRC claimed that up to one in every two girls abducted by the Lord's Resistance Army will return home HIV positive.
3. Queen's University, Harvard University and a US NGO called Discover the Journey have recently collaborated (March 2012) on a project to reduce the stigma experienced by formerly abducted children who had returned home after being captured by the rebel Lord's Resistance Army, currently operating in northern DR Congo.

Engineer Guy shows how a phone accelerometer works, knows what's up and sideways (video)
We love finding out how things work, and arguably one of the most important parts of the smartphones and tablets we thrive on is the accelerometer gauging our device's orientation. Imagine our delight, then, when we see the University of Illinois' Bill Hammack (i.e. The Engineer Guy) giving a visual rundown of how accelerometers work. Although it's certainly the Cliff's Notes version of what's going on in your Android phone or iPhone, the video does a great job of explaining the basic concepts behind three-axis motion sensing and goes on to illustrate how MEMS chips boil the idea down to the silicon form that's needed for our mobile hardware. Hammack contends that it's one of the coolest (and unsung) parts of a smartphone, and we'd definitely agree; you can see why in the clip after the break.
Engineer Guy shows how a phone accelerometer works, knows what's up and sideways (video) originally appeared on Engadget on Tue, 22 May 2012 20:06:00 EDT. Please see our terms for use of feeds.

5 Internet Marketing Musts for Small Businesses
You?ve done it. You have realised that the web is now another piece of your business puzzle and something you need to make a start on. Great!
But all of this can seem a little daunting with a lot of technical jargon and constant updates on ideas. However, this can be a little guide to get your started.
- First up, ?Stay Savvy? and get learning. SEO is your friend, as it?s a basic knowledge of HTML. Unfortunately, this area has become a playground of good cops and bad cops and I?ve heard of small businesses paying up to 865,000 rupee for Search Engine Optimisation (SEO) services which I totally over prices! There is no such thing as a free lunch and with internet marketing this is very true! By learning the basic elements of this yourself, you can begin some elementary optimisation yourself and understand what your consultant tells you. Follow your gut and seek some independent opinion on any quotes and strategies you might get. Google offer some great resources for beginners to find out about SEO and what makes great optimisation and On the whole, there are some amazing SEO?s out there but just be careful!
- Get Creative. Internet marketing has changed the link between businesses and customers and modern consumers are much more aware of the power they have. With this comes a change in your plans. If you spend 5 hours a week planning traditional, outbound marketing change that and dedicate your time to inbound marketing, where customers come to your website for a whole range of reasons! (see the great graphic above courtesy of Jennifer Sable Lopez at SEOmoz) This is also your time to get thinking. A great way of getting more traffic to your website is by creating quality, interesting and unique content. Infographics, videos, twitter trends, images, anything! This is your time to shine and make yourself better than your competition.
- As this might be something new for your business, find out what works for you and your customers early on. Try surveys and analytics and test your internet marketing. If ?x? doesn?t create and leads but ?y? does then test that. I suppose the best thing to do is not be caught up in any trends and make an internet marketing strategy that works for you and your customers; smiling faces all round.
- Remember your centre point is your website and your brand. Everything you do needs to lead to your website so ensure that the more you expand your online marketing make sure it ultimately leads back to your main site. The same goes for social media. If your Twitter followers are getting bigger then gently push them to your site. Inbound/Internet marketing works in a funnel style (see above) so the aim is to keep users retained from start to finish.
- Measure. Set up Google Analytics, record your social subscribers and email subscribers and make regular records of their numbers to understand your audience. Google tells you everything and there are measurement tools for Twitter too to understand your audience like Social Bro. Keep an eye on what works and adapt your strategy around that to keep your users following through the entire website.
So there you go. A basic introduction to internet marketing. Make a start and watch your website grow! Get your team together and decide what you are going to do and make some performance indicators. There is a lot of help online with blogs like HubSpot, SearchEngineLand and SEOmoz (to name a few) but the key is to attract users to your site, to convert them to customers and then create leads into your business. All of your internet marketing should focus on this 3 step funnel approach.

Chicago NATO Command and Communications Operations Update ...
Shortwave America
Shortwave America knew that the mainstream media was not telling Chicago residents everything, and that they were being good sheeple to appease the whims of the united States Federal Government. Chicagoans have been asking questions for months now about why the security boundaries were drawn the way they were and just how NATO is operating inside their city boundaries under the protection of a whole slew of U.S. federal protection.
Today, Shortwave America set out to get those questions answered. Today, the citizens of the world even beyond the borders of the city of Chicago know everything. Here is a PDF File from Chicago Police News Affairs regarding media access and related issues.
In the NATO Communications Guide and Live Communications Coverage post here at Shortwave America, we know which systems they are using and which other communications systems should be watched all while following NATO live from this publication.
There is now photographic evidence on hand to back up the story that is being told here. These new and yet untold details will help you follow NATO while they convene in Chicago.
Let?s start with the locations of the various Command and Communications Posts:
There are multiple sites starting at Navy Pier (Command and Communications site #1). The Hollyhock is stationed there, and her transponder HAS INDEED been turned off. At Navy Pier on the secure side of the Chicago Locks operations area is the U.S. Coast Guard Command and Communications operation.
The next location (#2) is the Chicago Police Department boat house next to the Chicago Locks. In the photos below, we see the Coast Guard patrolling the locks and river side of the operation for NATO, and the Chicago Police boat house itself, along with some of the marine support vessels to include the Chicago Fire Department as always.
NATO Command and Communications location number three is the Chicago Museum Campus itself. The greater compliment of operations are based out of the Chicago Field Museum. In the photos below, we see the U.S. Secret Service, various other government agencies and also various consulates operating at the Field Museum.
NATO Command and Communications operation number 4 is at the Burnham Harbor parking lot just across the way from Northerly Island and adjacent to the old McCormick Place. In the photos below, we see various members of the military / Homeland Security guarding the operation, meeting with superiors. We also see the greater cross section of the operation to include their communications and surveillance equipment which includes cellular and microwave frequency operation.
Next up is aviation and ground support operations! Many Chicago residents have witnessed the U.S. Coast Guard aggressively patrolling the skies, along with military and other helicopters. Shortwave America has not only photos, but the exact details on head of state and dignitary transportation operations. Other information includes that which is already publicly available data on the Homeland Security helicopters for all of you aviation enthusiasts.
Before we get to the photos, here is how the dignitary / head of state transportation is working! The military and Homeland Security helicopters meet the honored guests at area airports. Those airports are O?Hare, Chicago Executive, Rockford, and one other airport that is unconfirmed. They then fly the guests to Northerly Island (used to be Meigs Field) at Burnham Harbor, where they are are then transported under heavily armed guard using U.S. Government buses and unmarked vans to the old McCormick Place. They are then taken into and through that building to the new McCormick Place and they are also taken DIRECTLY to the NEW McCormick Place building thus bypassing the old McCormick Place and escorted into an entrance just to the North of the public entrance. (all of this was personally observed). This can also be confirmed by way of the NATO Security Transportation Plan PDF file at the Shortwave America NATO Communications Guide and Live Communications Coverage post.
Information above explains why they drew the street closures the way they did. Get it now? Museum Campus being used for operations, Burnham Harbor being used for operations, Northerly Island being used for operations, Lake Shore Drive and Balbo is staging for Chicago Streets and Sanitation equipment, etc.
The reverse order will be taken when NATO guests leave.
Here are the photos!!
A search of the U.S. FAA Aircraft Registration System revealed the information contained in the screen shots below:
As I started further south to see what else I might find, this reporter crossed through the Chicago Police Gold Star Families Park, the Chicago Fire Department Memorial Park, and the walk was VERY surreal because you could FEEL the tension in the air as joggers and cyclists watchfully went about their daily exercize. No one wanted to make a friendly greeting to other people, even a smile and wave were seen suspiciously. At Lakeshore Drive and 31st street, I met up with a photog from the Chicago Tribune who told me that even the Trib was having trouble identifying the aircraft and determining their operations in regard to NATO. After seeing my photos, the photog called his office and told them my findings. Let?s just say it was a very short conversation.
One important observation made through this day long trek was 7 to 8 foot high steel grate fences linked together were seen almost everywhere when it came to major roadsides and overpasses. The fence sections are approximately three to four feet across.
The greater amount of Federal resource and Law Enforcement presence was observed at the various Chicago Federal Buildings. What had previously been just a few vehicles here and there were now street lengths deep and even the local side streets / alleyways near the Federal buildings were packed with Federal Protective Service / Homeland Security Police vehicles.
This reporter did speak to Homeland Security Officers who state that they expect the city to absolutely erupt in never before seen violence over the weekend. The officers spoke on condition of anonymity because they were not authorized to speak to the press. While speaking with the officers, it was observed that their radio transmissions are in the clear as of this time.
One other thing that is proven by this report: Taxpayers are INDEED on the hook for this NATO fiasco! All of this costs money, and it isn?t coming from thin air! Hey, Rahmbo and Obummer?.where are the funds coming from? WE WANT TO KNOW!!!!
Thanks to all of you for choosing Shortwave America as your up to date source for NATO 2012!

Easy exterior projects that add lasting beauty to your home | U.S. ...
The easy patio fix that's eco-friendly
Adding pavers to any outdoor area can transform cracked slabs or troubled grass areas into virtually instantaneous living space. But most do-it-yourselfers shy away from working with heavy concrete or brick. Andy Vander Woude of AZEK VAST Pavers says you can install a new patio, walkway or driveway with new pavers that install three times faster on a unique grid system over a traditional sub-base. At less than half the weight of concrete, they contain up to 95 percent recycled scrap tires and plastics. "There is no need for wet saws or heavy equipment," Vander Woude said. "You can also resurface an old patio, deck or flat roof without the sub-base preparation - just assemble the grid and lay the pavers in like a big puzzle." He added, "It's a way to create instant living space that respects the planet."
Add curb appeal for lasting first impressions
You can increase curb appeal with stylish and functional, quality amenities that dress up an entryway, yard, patio, porch or deck. According to the experts at Walpole Woodworkers, known for outdoor furnishings handcrafted in Maine since 1933, consider classic accents such as lantern and mailbox posts, pergolas, arbors, window boxes, deck planters and birdhouses handcrafted from new advanced materials, with long-lasting benefits and low-maintenance. Walpole offers these and many other products handcrafted in modern cellular PVC. This advanced material looks just like natural wood, but won't rot, split, or warp and will maintain its good looks for years, virtually maintenance free. Call 1-800-343-6948 for a free catalog.
Light up the night
With the emergence of inspired outdoor living spaces and new trends in materials, it only makes sense that homeowners search for a way to enjoy their deck parties well into the night. Deck rail lighting has become extremely popular, with LEDs the preferred way to create ambient lighting. Michael Gori, Director of Product Management at AZEK Building Products, makers of AZEK Deck, said lighted post caps and light strips concealed under the hand rail are in high demand. "The lighted post caps add an elegant touch, while the hand rail lighting casts a downward glow, reducing hot spots and attraction to insects," he said. "The combination of the two light sources adds a dramatic effect for nighttime entertaining." Gori adds, "With warm, ambient lighting, there's no longer a need to head indoors when the sun goes down."
Know when to paint caulk
Did you ever poke your finger at a caulk bead to see if it's dry and ruin your nice work? "Caulk needs to dry for about one to two hours, before it can be painted-whether you're working indoors or out," says Jason Ringling, marketing director at Red Devil, a company with a 140-year old history in the industry. "Painting too soon can cause sagging and cracking and then the entire job has to be re-done." He says that there is a new technology called ColorCure sealant that turns from pink to white when it's ready to paint, so there's no guesswork. And if the job gets interrupted, you know exactly where you left off. "Formulations are getting smarter to help you work more efficiently," he adds.
When fixing up or adding beautifying elements to your home exterior, be sure to investigate all the options--from new formulations in caulks and sealants to alternatives to traditional materials that offer easy installation and better performance over time.

UN nuclear chief in Iran on key mission
TEHRAN, Iran (AP) ? The head of the U.N nuclear watchdog met Monday with Iranian leaders in Tehran on a key mission that could lead to the resumption of probes on whether Iran has secretly worked on a nuclear weapon.
Yukiya Amano of the International Atomic Energy Agency was quoted by Iranian state television as saying that he held talks with Iranian officials in a "good atmosphere."
The one-day visit ? Amano's first since becoming the IAEA chief in 2009 ? is focused on getting Iran to agree to terms that will allow IAEA probes of suspect Iranian sites, including the Parchin military complex where the agency had reported suspicious activities in the past.
Tehran denies having worked on atomic weapons, saying that its nuclear program is for peaceful purposes like research and cancer treatment. It says Parchin is only a conventional weapons site.
Neither side made mention of Parchin in remarks to the press after initial negotiations, keeping their statements general. Amano also said both sides discussed "various issues including nuclear disarmament, peaceful use of nuclear energy and future actions."
Top Iranian negotiator Saeed Jalili meanwhile said that he had "good talks with Amano in this regards and, God willing, we will have good cooperation."
Inspecting Parchin, southeast of the capital Tehran, was a key request made by senior IAEA teams that visited Tehran in January and February. Iran rebuffed those demands at the time.
But with both Iran and the IAEA reporting progress in a previous round of talks last week, anticipation ahead of the visit was high. While expressing some optimism, Amano said he could not predict whether he would clinch a deal that would allow his agency to renew its long-stalled probe.
Amano's trip is significant both for what it can achieve in terms of probing Iran's secretive nuclear program and as a mood-setter for talks Wednesday in Baghdad between Iran and the United States, Russia, China, Britain, France and Germany.
The six world powers are at the forefront of trying to persuade Tehran to curb its nuclear program and ease concerns it wants to use it to make nuclear weapons. For its part, Iran will seek to stay looming U.S. and European Union sanctions on its oil exports at the Baghdad talks.
The six will attempt to get Iran to commit to stop enriching uranium to a level that can be turned quickly into the fissile core of nuclear warheads, while ignoring ? for now? its program of lower enrichment, which would take longer to turn toward weapons-making.
Iran insists it is enriching uranium only to produce nuclear fuel and for cancer treatment. It denies that it worked secretly on developing components of a nuclear arms program.
Parchin is especially significant since the IAEA believes Iran in 2003 ran explosive tests needed to set off a nuclear charge. The suspected blasts took place inside a pressure chamber. Iran has never said whether the chamber existed.
As Amano arrived, Iranian lawmaker Heshmatollah Falahtpisheh told The Associated Press that Tehran will likely accept more inspections of Parchin, "if it feels there is good will within the (IAEA) agency."
But Falahtpisheh, a member of the influential parliamentary committee of national security and foreign policy, warned that this new openness will likely come with expectations that the West would in return ease international sanctions on Iran.
"In opening up to more inspections, Iran aims at lowering the crisis over its nuclear case," said Falahtpisheh. "But if the sanctions continue, Iran would stop this."
A political analyst in Tehran, Hamid Reza Shokouhi, said Iran is carefully watching to see if the West shows more "flexibility and pays attention to Iranian demands" during Amano's trip.
"Then Iran will show flexibility, too," said Shokouhi.
Both sides appeared to be doing their best to dispel tension that had built up between Tehran and the agency in the past. Amano expressed regret for the assassination of four Iranian nuclear experts over the past two years and said, "I will do my best to protect classified information" of IAEA member countries.
Iran blames the killings on Western and Israeli intelligence and has in the past accused the agency of leaking information on its nuclear program as well as its nuclear scientists. The IAEA denies this.
Jalili meanwhile praised the IAEA for what it said was its work "opposing false claims of the U.S. in Iraq" regarding Baghdad's alleged possession of weapon of mass destruction prior to the 2003 U.S.-led invasion, according to the official IRNA news agency.
Amano also met Fereidoun Abbasi, head of Iran's nuclear agency, as well as Foreign Minister Ali Akbar Salehi during his visit.

Scientists Discover Marker to Identify, Attack Breast Cancer Stem ...
HOUSTON ? Breast cancer stem cells wear a cell surface protein that is part nametag and part bull?s eye, identifying them as potent tumor-generating cells and flagging their vulnerability to a drug, researchers at The University of Texas MD Anderson Cancer Center report online in Journal of Clinical Investigation.
?We?ve discovered a single marker for breast cancer stem cells and also found that it?s targetable with a small molecule drug that inhibits an enzyme crucial to its synthesis,? said co-senior author Michael Andreeff, M.D., Ph.D., professor in MD Anderson?s Departments of Leukemia and Stem Cell Transplantation and Cellular Therapy.
Andreeff and colleagues are refining the drug as a potential targeted therapy for breast cancer stem cells, which are thought to be crucial to therapy resistance, disease progression and spread to other organs.
?It?s been difficult to identify cancer stem cells in solid tumors,? Andreeff said. ?And nobody has managed to target these cells very well.?
The marker is the cell surface protein ganglioside GD2. The drug is triptolide, an experimental drug that Andreeff has used in preclinical leukemia research. The team found triptolide blocks expression of GD3 synthase, which is essential to GD2production.
Triptolide stymied cancer growth in cell line experiments and resulted in smaller tumors and prolonged survival in mouse experiments. Drug development for human trials probably will take several years.
Cancer stem cells are similar to normal stem cells
Research in several types of cancer has shown cancer stem cells are a small subpopulation of cancer cells that are capable of long-term self-renewal and generation of new tumors. More recent research shows they resist treatment and promote metastasis.
Cancer stem cells are similar to normal stem cells that renew specialized tissues. The breast cancer findings grew out of Andreeff?s long-term research in mesenchymal stem cells, which can divide into one copy of themselves and one differentiated copy of a bone, muscle, fat or cartilage cell.
Andreeff has shown these mobile mesenchymal stem cells home to wounds, including tumors, making them potential carriers of cancer therapy.
An important cellular transition also comes into play.
Co-senior author Sendurai Mani, Ph.D., assistant professor in MD Anderson?s Department of Molecular Pathology and Co-Director of the Metastasis Research Center, is an expert on epithelial-to-mesenchymal transition (EMT). About 85 percent of all solid tumors start in the lining of an organ, called the epithelium. Mani and colleagues at MIT showed that epithelial cells can be induced to take on stem cell properties by forcing them to undergo EMT.
?This change from stationary epithelial cells to the mobile mesenchymal stem cells is an important step in metastasis,? Mani said.
Andreeff and Mani in 2010 discovered that human mammary epithelial cells that undergo epithelial-to-mesenchymal transition act similarly to human bone-marrow-derived mesenchymal stem cells. They can home in to wounds and differentiate into the same cell types.
GD2 separates cancer stem cells from other tumor cells
In the current project, the researchers hypothesized that the cell markers expressed on the surface of mesenchymal stem cells would also be expressed on the surface of breast cancer stem cells.
They found that GD2 expression, one such mesenchymal stem cell marker, divided the breast cancer cell lines into two distinct groups: about 4.5 percent of cells were GD2-positive and about 92.7 percent were GD2-negative.
GD2-positive breast cancer cells:
- Form twice as many mammospheres, a clumping of cells considered an indicator of tumor-forming capacity, as compared to GD2-negative cells. And the spheres were three times as large.
- Migrate four times as fast as GD2-negative cells.
- Form five times as many tumors when 10 cells of each type are transplanted into mice.
?
GD2-positive cells also have general cancer stem cell marker
A known combination marker of cancer stem cells is high expression of CD44 and low expression of CD24 surface proteins. The researchers found 85 percent of GD2-positive breast cancer cells were CD44 high/CD24 low, while only 1 percent of GD2-negative cells shared that characteristic.
An analysis of 12 human breast cancer tumors found an even higher correlation of 95.5 percent between GD2+ cells and CD44 high/CD24 low status.
Comparing gene expression between GD2+ cells and CD44 high/CD24 low cells revealed 100 percent correlation in the expression of 231 genes.
GD2+ cells had greater expression of genes involved in migration, invasion and epithelial-mesenchymal transition than GD2- cells. They also had a nine-fold increase in GD3 synthase, a key enzyme in the eventual synthesis of GD2.
Further experiments showed that:
- Inducing EMT raised the percentage of GD2+ cells in two breast cancer cell lines.
- Knocking down GD3 synthase cut the percentage of GD2+ cells by more than half.
- Mice injected with 1 million breast cancer cells having a small interfering RNA that blocked GD3 synthase never developed tumors even after eight weeks, while all of the control mice with active GD3S developed tumors.
?
Triptolide stymies tumor growth, extends survival
The researchers then used triptolide, a known inhibitor of GD3 synthase, to treat immune-deficient mice injected with breast cancer cells. Of the mice treated, 50 percent did not develop breast cancer and the other half had smaller tumors than the control mice. The treated mice also lived longer than the controls.
GD2?s function in cancer stem cells remains unclear. ?As GD2 is an immune suppressant, it would be needed by cancer stem cells to counter immune cells during metastases,? said first author Venkata Lokesh Battula, Ph.D., of MD Anderson?s Department of Leukemia. ?Inhibition of GD2 expression in cancer cells may enhance the inherent ability of immune cells to kill cancer cells.?
